Normally you would just have a cell/column where QTY in stock was inputted, maybe F2 then hide it and then C2 is "=F2-E2".
We could give you a more complicated solution but might as well keep it simple.

(the complication problem is having a cell being both the place you input a value and the place that displays the new value)
