Well, there are a few ways to do this...
By default, all cells in Excel are set as 'Locked', but it has no effect unless you Protect the Sheet (REVIEW tab >> PROTECT SHEET).
If you only want users to be able to access certain cells, select those cells, then press Ctrl + 1. That will open the Format Cells dialog. On the protection tab, remove the check next to 'Locked' so those cells become unlocked and editable. Then protect the sheet (password is optional).
The opposite side of that is you could select the whole sheet (by clicking on the triangle above the 1, left of the A) press Ctrl + 1, then remove the check to unlock every cell.
Then select all of the cells you need to have locked, press Ctrl + 1, and click to add the check back in the 'Locked' checkbox. Then, when you protect the sheet, the sheet will be full of unlocked cells, except for those that you locked.

you "unlock all non-yellow cells / blue cells" first, then you lock entire worksheet..
