Try applying the following CF rule to range B2:AF5000:
=AND($A2<>"",B2="")
Like this:
=AND($A2<>"",B2="")

by the way, why you added "$" only to A2?
If you didn't do that, then - when it looks in column B, it would look for a non-blank in B. The $ anchors it so that the first condition is always sought in column A, the second condition is sought in each individual cell.
